Abstract: Small molecules can be used to probe biological states of bacteria to reveal essential functions required for survival. This approach can be particularly powerful in studying Mycobacterium tuberculosis due to the variable physiologic states adapted by the bacilli within the infected human host and the challenge of classical genetic approaches in this system. We will discuss approaches that we are taking to understand the physiologic states adopted by TB that may contribute to its refractoriness to drug therapy, termed drug tolerance, and the insights that can be gained that may guide more effective drug discovery |
